背景
粒细胞-巨噬细胞集落刺激因子(GM-CSF),也称为集落刺激因子2(CSF2),是单体糖蛋白,不同于粒细胞集落刺激因子,它特别促进嗜中性粒细胞的增殖和成熟,GM-CSF影响更多的细胞类型,特别是巨噬细胞和嗜酸性粒细胞。在类风湿性关节炎的关节中发现GM-CSF含量很高,而将GM-CSF作为生物学靶标可以降低炎症或损害。在重症患者中,GM-CSF已被试用为一种重症免疫抑制疗法,并已显示出有望恢复单核细胞和中性粒细胞的功能。GM-CSF的功能有:骨髓谱系细胞的骨髓产生和分化,肺泡巨噬细胞的发育和维持,招募和分化单核细胞来源的DC(包括产生T细胞的IL-23和TH17极化),常规的DC成熟和抗原呈递(包括皮肤和小肠中表达CD103的DC),M1巨噬细胞极化(包括促炎细胞因子的产生,吞噬作用,抗原呈递),中性粒细胞引发和激活(包括吞噬作用,氧化性爆发和一氧化氮生成)等。
