Background

Matrix metallopeptidases (MMPs) consists of 24 known human zinc proteases, it plays an important role in breaking down components of the extracellular matrix (ECM). As a protease, it has been found the substrates of MMP include cytokines, chemokines, growth factors, binding proteins, cell/cell adhesion molecules and other proteases. Except for a very few MMPs, most of the them in the whole MMPs family shares common structural motifs, including a pro-peptide domain, a catalytic domain, a hinge region and a hemopexin-like domain.

Generally, most of MMPs are secreted to inactive pro-enzymes form before they are involved in the activity of enzyme metabolism. Usually, MMP is involved in the modulation of enzyme activities on the following several levels, including transcription, pro-enzyme activation, endogenous inhibitors, and tissue inhibitors of metalloproteinases (TIMPs).
