E.coli HCP 产品性能对比分析

背景
纵观生物药物生产的历史,从药物终产物中,有效地去除和检测HCP杂质,有着大量的研究和发现。这些杂质的存在,对临床病人的安全和药物稳定性有着广泛的影响。
目前,常见的蛋白药物表达系统,有原核表达系统,如Escherichia coli ,E.coli;植物表达系统;真菌表达系统,如毕赤酵母,酿酒酵母;昆虫表达系统,如Spodoptera frugiperda cells,SF9昆虫细胞;哺乳动物细胞表达系统,如中国仓鼠卵巢细胞(Chinese hamster ovary cells, CHO),非洲绿猴肾细胞(African green monkey kidney cells,Vero)等。其中哺乳动物细胞表达系统,在临床药物的生产中,使用较为广泛,但是价格昂贵。产量最高,价格低廉,用途最广泛的是E.coli表达系统。
E.coli表达系统是目前使用领域最多元的表达系统,由于其培养简单,转化效率高,成本低,产量高等特点,而被广泛应用。诸如不需要修饰的人胰岛素和生长激素,同时自然状态下需要糖基化修饰,但可以不糖基化修饰的蛋白,如 α-,β-和 γ-干扰素,以及白介素2和肿瘤坏死因子,仍具有生物学活性[1,2]。
对HCP的检测和鉴定方法,通常的金标准是ELISA和LC-MS法。其中,前者可以高通量定量,且价格低廉,后者可以定量定性,但是价格昂贵,且通量较低。两者需要正交搭配使用,来对药物中的HCP进行定性和定量。
有较多文献报道,不同药物的相同纯化工艺,或者相同药物不同工艺的HCP残留均有较大差别,因此,国内外的药物HCP定量结果不尽相同[3]。
E.coli HCP与CHO HCP有较大的差别
大致可以分为以下几类:
1、黏附素(Adhesins):Curli (CsgA) ,形成淀粉样纤维,促进生物膜形成;
2、毒素(Toxins):α-溶血素(HlyA), 孔形成毒素,溶解红细胞和上皮细胞;
3、侵袭素(Invasins):OmpA, 促进E. coli侵袭血脑屏障(NMEC),抵抗补体杀伤;
4、免疫逃逸相关蛋白: LPS(脂多糖), 诱导炎症,但逃避免疫识别;
5、分泌系统效应蛋白:EspC(自转运蛋白酶),破坏上皮屏障;
6、宿主互作效应蛋白:NleH, 宿主NF-κB通路,减少炎症;
7、噬菌体/质粒编码毒力蛋白:Colibactin, 基因毒素,诱导DNA损伤。
Jones等报道,残留的HCP不仅会有免疫原性,而且还有药物辅料降解的功能。对宿主菌/细胞高风险因子基因做基因敲除,或使用空载HCP模拟药物生产和纯化流程,如深层过滤、亲和纯化、阴阳离子纯化产物中的HCP残留分析尤为重要。在优化药物优化纯化过程中,不乏有较难去除的HCP。Zhu等报道过,E.coli表达的malaria vaccine在纯化过程中的HCP含量范围在90 ng or <1100 ppm/剂。但是临床能接受不形成慢性疾病的HCP含量范围仅仅<100 ppm/剂[4,5,6]。
所以,对HCP需要应检尽检,防止漏检,不断提升检测灵敏度和覆盖度。
